High-Performance Liquid Chromatography (HPLC) of Propionic Acid
The core instrument for this method is the high-performance liquid chromatograph. The instrument must also be calibrated, with an appropriate column and mobile phase selected. For propionic acid detection, a C18 reversed-phase column is commonly used. The mobile phase is typically a mixture of water and an organic solvent (such as methanol) in a fixed proportion, with an appropriate buffer salt (e.g., potassium dihydrogen phosphate) added to adjust the pH and ensure good separation. Taking the detection of propionic acid in cosmetics as an example, the cosmetic sample is first pre-treated (e.g., diluted, filtered) to remove impurities that may interfere with detection. The treated sample is then injected into the high-performance liquid chromatograph. During operation, the mobile phase carries the sample through the column, where propionic acid is separated from other components through multiple partitioning between the stationary and mobile phases. A UV-Vis detector can be used, as propionic acid absorbs light at specific wavelengths. By measuring the absorbance of propionic acid at the specific wavelength and comparing it with a standard curve, the propionic acid content in the sample can be determined. The standard curve is constructed by analyzing a series of propionic acid standard solutions of known concentrations, with concentration on the x-axis and absorbance on the y-axis.
