After optimization, a typical set of determination conditions is provided below. Analysts may fine-tune these based on the performance of the actual instrument and chromatographic column:
Sodium Formate Mobile Phase: 20 mM potassium dihydrogen phosphate buffer (pH 3.0) : methanol = 95:5 (v/v). The buffer must be filtered through a 0.45 µm membrane filter and degassed. The pH significantly affects the peak shape and retention time of sodium formate.
Chromatographic Column: Reversed-phase C18 column, 250 mm × 4.6 mm, 5 µm. Ensure the column is in good condition with acceptable efficiency.
Flow Rate: 1.0 mL/min (constant flow).
Sodium Formate Column Temperature: 30°C, maintained within the 25–40°C range for analytical stability.
Detection Wavelength: 210 nm. Sodium formate exhibits end absorption near this wavelength; ensure the detector’s deuterium lamp has sufficient energy.
